In this particular case, tolerance comes from something called a 5-HT2A receptor. If you don’t remember biology class, then don’t worry—we’ll sum it up. Basically, on many cells—including neurons that make up the brain—are receptors. These are small proteins that often stick outside of the neurons and react to various biochemical signals that attach to them.However, not everything that passes by can interact and attach with these receptors. Usually, only a few particular chemicals can activate any given receptor.

For example, 5-HT2A is a serotonin (aka, 5-HT) receptor. That means that serotonin (5-HT) and other serotonin-like chemicals can activate them.

When these receptors are stimulated, they become inactive for a period of time. It’s also thought that the neurons can sense that the 5-HT2A receptors are being activated too much and stop sticking them out.

That means that even if you took more psilocybin, there are fewer receptors for them to attach to, leading to less effect. This is why we need to take at least 2 days off a week when microdosing and now we can use receptor cleanse to help replenish, reset and prepare the receptors for the following week. This also allows the microdosing effects to still work on the days off and balance the brain over time for the microdosing to work more efficiently (up to 30%).
